I've used WP for years, but am just straying into WP-MU installs for various jobs.
Clients have asked me and I wanted to find out - is there a way to create large amounts of user accounts and/or blogs associated with them, en masse? Maybe an automated method?

You do realize that you just asked us how to create splogs, right? :)
Not doubting you (Checking out your profile shows this probably isn't your intent) but I just wanted to point that out.
edit: Give this plugin/ script a look. You can probably go from there. Googling for 'create many blog' may work as well:

Sorry yes it's not for that purpose! It's for marketing purposes, research companies, who need ways of creating large amounts of small blogs.
