Does anyone know of a way that we can run WordPress MU to support blogs for our employees, but not allow just anyone to create one?

Best bet would be to use the Limit the email registrations feature at Dashboard -> Site Admin -> options and set it to only your companies email domain.

That and remove the all of the links to the signup page as well even getting rid of the signup file.

You could also accomplish a lot of this by restricting access to your wp-signup.php file using .htaccess. With some cleaver use of Apache features you could easily restrict access to those pages from non-corporate networks clients.
