As a global site WPMU administrator, I should be able to see every new user that registers in my site. Is this how it should work? As of now, I'm not able to see new register users on the admin site. Do I need to install a plugin for this functionality?

Under Site Admin -> Blogs for the blog and Users for users.
Yes, you see them all by default. Those menus list them all.

You're not logged in as the Site Admin. The "admin" username...
If you are logged in as "admin", make sure you're running WPMU and not single WP.
I'm logged in as Admin, but it looks like this is a known issue, here is the post I found, and you participated in it about two years ago.

It's not a known issue. I've installed new copies of WPMU at least half a dozen times in the past week and it works for me. The codebase has changed significantly in the past two years.
- is this a fresh install?
- did you change anything about the admin user?
- are you logged in to the main site?
Try this: yourdomain.com/wp-admin/wpmu-blogs.php
Substitute yourdomain.com for your actual domain name.
very strange, I get the message "You don't have permission to access this page". However, I'm logged in as admin, maybe the site broke.
any other suggestion?

That may be a possibility, but I don't know what I couldn't have changed. is there a way to restore it?

yes it is, and I'm logged in to the main site. I logged in in the form; mysite.com/wp-admin. the only plugin I'm using for now is an SMTP pluggin to send automatic emails to registered users.

Forgot to mention, I'm still testing this site and WPMU functionalities. I just want to make sure that I won't get this type of issues on my production site.
