I been doing research on building a website using multiple WordpressMU.

    Example:
    Main site: mydomain.com
    Subdomain: sub1.mydomain.com
    Subdomain: sub2.mydomain.com
    Subdomain: sub3.mydomain.com
    Subdomain: sub4.mydomain.com

I wanted to know if this could be done with the following features:

    1 database
Mods and plugins work for all subdomain
Seperate admin and managing per subdomain without accessing other subdomain
Single login for all users thru main domain
Users be able to post blog in any subdomain

What issue would i run into when theres an update or new mod plugin setup install? Any help or ideas would gladly be appreciated, thanks.

What you're describing is just basically what a single installation of WPMU can already do using just one database. And yes, you can install it in subdomain structure like you wanted above and subdirectory structure.

    - 1 database --- yes and can also expand to multiple databases
    - Mods and plugins work for all subdomain --- of course
    - Seperate admin and managing per subdomain without accessing other subdomain --- sure, that's what it is originally set up for
    - Single login for all users thru main domain --- sure, that can be done, or they can log into their own blogs directly as well
    - Users be able to post blog in any subdomain --- yes, just set up your WPMU as you want it to be

    Before installation, please read
    http://trac.mu.wordpress.org/browser/trunk/README.txt
    http://codex.wordpress.org/WordPressMU
    http://codex.wordpress.org/Installing_WPMU

Backing up database and server files before upgrading has always been de rigueur for any installation. Of course, it is always good to check plugin compatibility vis-a-vis new WPMU version, but unless there have been major changes in codebase, no problems in upgrading plugins at all. Of course, there will sometimes be hiccups along the way. Just backup, backup and backup :-)

When i do updates and plugins, do i have to update all subdomain or just the main domain? Thank you for your help.

You just replace the single copy of the code and if its a core WPMU update there is an option to update all blogs.
