Does the server have to have wild cards enabled, to use sub-domains as my hosting company states it will only make changes if I can give them something definite to work on, as they don't support third party scripts,
ie sub-domain myblog.mysite.com.

Yes, if you want to use subdomains, you must have wildcard subdomains enabled.
There are instructions in the readme.txt file that explains it if you have trouble explaining it to your hosting company.

Gotta admit though that sounds like a poor answer from your host as it;s a setting on their end that needs to be changed. You're not asking for assistance with a script.
