I know I can edit these settings in wp-includes/wpmu-functions.php, but I'd like to avoid hacking the core files for convenience sake in future upgrades.
My questions are:
1) Is there a plugin that currently allows you to change the content of the first post/comment and the name of the first "commenter" (ie. Not Mr Wordpress).
2) If not (my searches haven't found any), would it be difficult to create a plugin to do this? I think this would be beneficial for most MU users, so if it's not too tought I might try to do it.

Ummm...I did post a suggestion there, but why would I ask a question there? and if you read my post...I did search the forum and nothing came up regarding an actual plugin.
For what it's worth, I have the plugin that is mentioned in the thread you linked to, but the version that was just released doesn't include what I'm looking for.
And like I said, if you actually read that thread, I did request it, but it wasn't something that ended up being implemented into it.
I think PerS is pointing out that opening yet another thread on a previously discussed subject is usually frowned upon with most forums.
You can set your First Post at Dashboard -> Site Admin -> Options -> It's on that page somewhere. You don't have to hardcode the file.
